Overview
Lead role in the design of protein expression constructs and maintenance of a database of constructs and protein inventory, including the purification and characterization of proteins (intracellular, extracellular, and multi-pass membrane) for our small molecule discovery program and the design and development of high content imaging cellular assays.
Responsibilities:Communicate protocols and manage workflows of external FTEs. Utilize various methods for biophysical characterization (SPR, DSF, DLS, TM-FSEC) of recombinant proteins samples for structural studies including CryoEM and crystallography. Project manage protein chemistry resources across multiple high-profile projects. Design and execute purification of native proteins from serum, tissue, and other biofluids. Lead troubleshooting efforts for challenging protein expression and purification processes. Analyze purified proteins using a range of protein characterization methods (including HPLC, SEC-MALS, DLS, SDS/PAGE and others). Establish methods to characterize and QC proteins for small molecule/protein binding.
Qualifications
Must have a master's degree in biology, biotechnology, related or the equivalent and 3 years of experience in biotech/biopharma, including with: Hands-on experience in protein purification from bacterial expression systems; Chromatographic techniques including the Akta FPLC platform and Agilent and Thermo Fisher HPLC systems; and, Culture and express recombinant proteins in e. coli. Of the 3 years of experience, 1 year of experience setting up and analyzing experiments from NanoDSF and Biacore instruments.
